小鼠同种抗原(Mouse Alloantigens)
同种抗原(alloantigen)是指同种动物不同的个体出现机体构成物质的抗原在遗传上的变异。主要组织相容性抗原(MHC)便是一类同种异体抗原,是引起同种异体移植免疫排斥反应的主要原因。MHC一般可分为三类:MHC class I、class II 和 class III. Mouse MHC抗原又叫作H-2抗原,此基因位于小鼠的17号染色体上。
Mouse MHC抗原由11个亚类组成:
1. 经典的MHC class I(“classical MHC class I”)抗原,即MHC-Ia抗原,包括H-2D、H-2K 和 H-2L三个亚类;非经典的MHC class I (“non-classical MHC class I”),又名MHC-Ib抗原,由H-2Q、 H-2M 和 H-2T三个亚类组成。
2. 经典的MHC class II(classical MHC class II”)抗原,即MHC-Iia抗原,包括H-2A(I-A)、H-2E(I-E)和d H-2P三个亚类;非经典的MHC class II (“non-classical MHC class II”),即MHC-Iib抗原,由H-2DM和H-2DO两亚类组成。
MHC class I分子由一个45 kD的高度糖基化的重链和一个12kD的β2微球蛋白通过非共价结合构成。Mouse MHC class II基因位于H-2 I 域,由一个33 kD的α链一个28 kD的β链构成。MHC class I表达于大部分的有核细胞上,在自身修饰细胞抗原(病毒感染或肿瘤细胞)呈递给CD8+毒性T细胞的过程中发挥重要作用。MHC class II抗原主要表达于抗原呈递细胞(B细胞、单核/巨噬细胞、DC细胞、Langerhans细胞等)和部分T细胞,主要功能是把加工过的多肽抗原呈递给CD4+细胞。
MHC分子具高度多态性,实验用小鼠常为天然纯合子个体,因而他们往往具有独特的MHC单元型(haplotype)。科学界把这些单元型分别用一些小些字母如a、b、d、k、q、s等来表示。例如Balb/c鼠的MHC抗原为H-2Kd、H-2Dd、H-2Ld、I-Ad和I-Ed。
